Can you activate package wide targets in the knowledge base?

Yes, you can automatically activate an entire collection (such as Academic Search Premier, where your library subscribes to the entire title list). For collections where your library only subscribes to selected titles, you can activate titles individually, either via web-based check boxes or using the title list upload tool.
